The first and most important text of the collection is an abbreviated form of the Rule of Saint Benedict, which governed life in the monasteries of the Benedictine order to which the mixed, but mainly feminine, convent of San Lorenzo in Venice belonged. In the title, the scribe, Gerolamo Zio, affirms that the text was requested by the Abbess Cipriana Michiel and, in the colophon (i.e. the final note), that he wrote it in 1560 (October).

Title: top of f. 1r, rubricated Regola del santissimo padre nostro messer san Benedetto Abbate: Abbreviata da patre Gieronymo Zio: de ordine della Reverenda Cipriana Michiel Abbadessa di questo Monasterio.

Prologue: starts on f. 1r. It is introduced by the rubricated words Prologo della Regola and it begins with the words Ascolta o figliuolo gli comandamenti del maestro; it ends on f. 1v with the words nel regno di colui, il quale ti ha chiamato.

Rule of Saint Benedict: starts on f. 2v with the words Delle generationi de Monachi. Capitolo primo (rubricated). Truovasi quatro sorte di Monachi; it ends on f. 22r with the words Et li regni di cieli saranno aperti a quelli, che le dette cose osservaranno. Amen.

Colophon: bottom of f. 22r, rubricated Fine della Regola di Santo Benedetto ridotta in capitoli brevi ad instantia della Reverenda Madonna Cipriana Michiel Abbadessa del Monasterio di Santo Lorenzo di Vinetia. Nel M.D.LX. del mese d'Ottobre.
